Summary:
The Ackert-Bicknell laboratory in the Colorado Program for Musculoskeletal Research, within the Department of Orthopedics is seeking to fill the position of Postdoctoral Fellow.
The Fellow will conduct research centered on the identification of fundamental mechanisms by which dietary fat impacts the physiology of the mesenchymal cells of bone and how this in turn affects development of the whole vertebrate skeleton.
An emphasis of this work will be the identification of gene by environment interactions that impact bone physiology.
This project involves the application of cutting-edge approaches in vivo and in vitro, including zebrafish, mouse and cell models combined with big data '-omics' approaches.
The results from this study are anticipated to substantially increase our understanding of when and how modifiable dietary factors impact skeletal development and will fill in gaps in knowledge left by other approaches such as Genome Wide Association Studies.
This research will be conducted in collaboration with Dr.
David Karasik at the Faculty of Medicine of Bar-Ilan University, Safed, Israel.
The postdoctoral fellow will be in constant communication with the Israeli counterpart to coordinate experiments being conducted as part of the larger research project.
This position is planned to last 2 years, with an option for an extended year.
Key
Responsibilities:
Zebrafish husbandry, genotyping, skeletal and non-skeletal tissue harvest.
Collection of skeletal imaging information from larval, juvenile and adult zebrafish and harvested tissue samples.
Generation of stable and chimeric transgenic zebrafish (so called CRISPANTS).
Collection of skeletal imaging information using live mice and harvested mouse tissue samples.
Molecular analyses of fish, mouse and human tissues, including Western blotting, qPCR/PCR, immunohistochemistry and immunofluorescence Culture of primary cells and established cell lines.
Transfection of cells with viral vectors or plasmid vectors.
Histology of musculoskeletal tissues (rodent and fish), including embedding, sectioning and imaging.
Collection of and analysis of RNAseq transcriptomic data.
Bioinformatic analysis using large data sets such as genomic, proteomic, lipidomic and transcriptomic data.

Qualifications:

Qualifications:
Applicants must meet minimum qualifications at the time of hire.
A PhD in Biology, Genetics, Biochemistry or related field.
Equivalent degrees may be considered.
At least three years of experience in Biological Research.
A strong publication record is required, including first authorship publications stemming from their previous research.
Preferred
Qualifications:

years of experience working with laboratory zebrafish and mice The CPMR has a funded T32 training grant and successful candidate will have the opportunity to compete for on the prestigious seats in this training program.
Therefore, preference will be given to those candidates eligible to receive NIH funded training support.
The successful candidate may have the opportunity to travel Dr.
Karasik's laboratory in Safed, Israel.
Eligibility to obtain a visa to travel to Israel is preferred, but not required.
Knowledge, Skills and Abilities:
Understanding of skeletal physiology and genetics Ability to work both in a team environment and independently Strong organizational and record-keeping skills Strong communication skills, both verbal and written Computer skills in both Mac and PC environments and use of standard programs such as Word and Excel Bioinformatics and or programming expertise in languages such as R and Python A strong understanding of Biostatistics.
